New tools for ubiquitin signaling.

چکیده
Protein ubiquitination is rapidly emerging as a key regulatory mechanism of many cellular processes. Important roles in proteasomal degradation are contrasted by various non-degradative functions of ubiquitin in intracellular trafficking, signal transduction and the DNA damage response. This versatility arises from eight structurally and functionally distinct polyubiquitin chains that co-exist in cells.1,2 Biochemical characterization identified proteins and enzymes that specifically assemble, recognize and hydrolyze polyubiquitin in a linkagespecific fashion, and structural analysis starts to reveal mechanisms of linkage recognition. What has been much more challenging is to understand the cellular roles of individual chain types and the interplay of the different forms of ubiquitination. A particularly fascinating area of ubiquitin research evolves around cytokine signaling pathways, for example, the response to tumor necrosis factor (TNF)-α. In addition to canonical roles of Lys48-linked chains as a degradation signal, the first described instances of non-degradative Lys63-linked ubiquitination in nuclear factor κB (NFκB) signaling more than 10 years ago opened the door for a vast number of discoveries. Recently, the picture became even more complex with reports implicating Lys11 chains and linear (or Met1linked) chains in TNFα signaling. The idea that linear chains cooperate with Lys63-linkages in activating cytokine-triggered kinases including inhibitor of κB kinase has resulted in some debate. However, the recent identification of SHARPIN as a component of the linear ubiquitin chain assembly complex, LUBAC, and its important roles in inflammation further supported that linear chains are key players in NFκB signaling. Despite this biochemical and genetic evidence, reagents that directly interrogate the roles of linear versus other polyubiquitin chains in vivo have been lacking. In this issue of Journal of Molecular Biology, Matsumoto et al. report the engineering of a linkage-specific antibody against linear ubiquitin chains and show its application in Western blotting, immunoprecipitation and immunofluorescence studies.12 A strategy that combined phage display

Ubiquitin-Activated Interaction Traps (UBAITs) identify E3 ligase binding partners.
We describe a new class of reagents for identifying substrates, adaptors, and regulators of HECT and RING E3s. UBAITs (Ubiquitin-Activated Interaction Traps) are E3-ubiquitin fusion proteins and, in an E1- and E2-dependent manner, the C-terminal ubiquitin moiety forms an amide linkage to proteins that interact with the E3, enabling covalent co-purification of the E3 with partner proteins. We de...
متن کاملThe Jak-Stat Signaling Pathway of Interferons System: Snapshots
Interferons (IFNs) are a family of small regulatory glycoproteins that play a central role in the defense against viral infections. Although IFNs have been initially discovered as antiviral factors, today they are known as an integral part of the cytokine network that affect a wide range of biological processes. IFNs exert their pleiotropic effects through their multisubunit cell surface recept...
متن کاملThe novel E3 ubiquitin ligase Tiul1 associates with TGIF to target Smad2 for degradation.
Ubiquitin-dependent degradation plays an important role in the negative regulation of TGF-beta signaling. Here, we identify Tiul1 (for TGIF interacting ubiquitin ligase 1), a novel E3 ubiquitin ligase that inhibits TGF-beta signaling by targeting both the activated receptor and Smad2 for degradation. Tiul1 associates constitutively with Smad7 and induces degradation of the activated type I rece...
متن کاملThe Colossus of ubiquitylation: decrypting a cellular code.
Ubiquitylation is an essential posttranslational modification that can regulate the stability, activity, and localization of thousands of proteins. The reversible attachment of ubiquitin as well as interpretation of the ubiquitin signal depends on dynamic protein networks that are challenging to analyze. In this perspective, we discuss tools of the trade that have recently been developed to dis...
متن کاملUbiquitin and its binding domains.
Post-translational modification by ubiquitin (ubiquitination, ubiquitylation, ubiquitinylation) is used as a robust signaling mechanism in a variety of processes that are essential for cell homeostasis. Its signaling specificity is conferred by the inherent dynamics of ubiquitin, the multivalency of ubiquitin chains, and its subcellular context, often defined by ubiquitin receptors and the subs...
